DNA Quantification Assay Kits FAQs
DNA Quantification Assay Kits - Frequently Asked Questions
Can the kits be used for both automated and manual quantification?
Yes. Both the 1× dsDNA HS Assay Kit and the ssDNA Assay Kit support automated quantification on compatible MGI platforms, including PrepALL, FLP, AIO and GenSIRO, as well as conventional manual workflows. Choose the workflow that best suits your sample throughput and testing needs.
Can the kits be used with Qubit™ fluorometers or fluorescence microplate readers?
Yes. Both kits support manual quantification on Qubit™ fluorometers and fluorescence microplate readers. For microplate readers, recommended excitation and emission wavelengths should follow the assay-specific protocol.
Does RNA affect ssDNA quantification?
It can, to a small degree. The dye in the ssDNA assay has a relatively low binding affinity for RNA, but RNA may still contribute to the signal and slightly overestimate ssDNA concentration. We therefore don’t recommend the ssDNA assay for samples containing significant amounts of RNA.
Does dsDNA contamination affect ssDNA quantification?
Yes. The dye in the ssDNA assay isn’t exclusive to ssDNA and also binds dsDNA, so dsDNA contamination can significantly affect results. If you suspect contamination, evaluate the sample separately using the 1× dsDNA HS Assay Kit.
Are two standards enough to generate a standard curve?
It depends on the instrument. On a Qubit™ fluorometer, use the two supplied standards as directed by the instrument workflow. On a fluorescence microplate reader, Standard 2 can be serially diluted in 1× TE buffer to build a multi-point curve and confirm linearity across the range you need.
