What makes the G99 a breakthrough in ultra-fast sequencing?

Launched on 9 September 9 2022, the G99 achieves super-fast sequencing through four key innovations: triangular configuration patterned-array with 600nm pitch-to-pitch distance for higher density spots, improved optical modules transcending light diffraction limits, innovative biochemistry achieving 2 minutes/cycle with 10s fluorescence reaction and 22s incubation, and rapid temperature control system with heating rates up to 10°C/s (1.5x faster than conventional PCR equipment).

For RUO models: G99RS (without bioinformatics module, outputs FASTQ files) and G99ARS (with built-in bioinformatics module supporting PFI, MetargetCOVID, MGAP, Flu track, FIS, and FIS SNP pipelines). For IVD models: G99 (NMPA and CE-IVD certified) and G99A (CE-IVD only). The system supports three flow cell types: FCS (40M reads), FCL (80M reads), and FCU (160M reads, launching June 2025).

The two flow cells (A and B) operate completely independently, allowing different read lengths and sequencing at different times without interference. Once one flow cell completes sequencing, it directly outputs FASTQ data without waiting for the other flow cell to finish. This dual mode enables concurrent or independent operation of multiple experiments with maximum flexibility.

G99 supports comprehensive reagent options: FCS flow cells with App-D PE150; FCL flow cells with SE100/PE50, App-D SE100, PE150, App-D PE150, App-D PE300, and SE400; FCU flow cells with App-D SE100, App-D PE150, and App-D PE300 (launching June 2025). Software upgrades are required: FCS corresponds to ECR3.3, FCU corresponds to ECR4.0. Most reagents contain formamide except SE100/PE50 configurations.

The system features three key convenience factors: pre-loaded reagent cartridge design with simple “press-to-load” operation eliminating manual liquid handling, optional built-in bioinformatics module enabling integrated sequencing and analysis, and intuitive user interface with guided icons, animations, and built-in instructional videos to prevent operational errors. The 2-in-1 sequencing and cleaning cartridge design automatically initiates cleaning after runs.

Yes, G99 supports Illumina adapters through integrated APP-D SE100 and APP-D PE150 sequencing sets that convert TruSeq and Nextera adapter libraries into compatible DNBs. Insert primers and barcode primers are incorporated within the sequencing sets without manual addition required. For dual barcode sequencing, no additional primer reagent kit purchase is needed as it’s built into the DNBSEQ-G99 sequencing sets.

G99’s algorithm is adapted from G400 ECR5.2, providing similar performance for unbalanced libraries as the G400 platform. The PE300 reagent on G99 differs from the G400 platform by not using MLG technology, offering alternative biochemical approaches for long-read applications.
