What makes the T1+ the flagship sequencer for high-throughput applications?
The T1+ is one of the world’s fastest mid-to-high-throughput genetic sequencers, producing PE150 sequencing data in as little as 24 hours (600Gb for a single flow cell and 1.2Tb for dual flow cells) with exceptional quality (Q40 > 90%). It features three types of flow cells (FCS, FCM, and FCL), offering flexibility to run one or two flow cells independently or simultaneously, with throughput ranging from 25-1200Gb to handle complex sequencing projects demanding speed and accuracy.
What configurations and models are available for the T1+?
The T1+ is available in two distinct versions: RUO (Research Use Only) launching globally in Q1 2025, and IVD (In Vitro Diagnostic) with CE-IVD certification expected by May 2025 and NMPA registration in Q2 2026. For RUO version, customers can choose T1+RS (without bioinformatics module, generates standardized fastQ reports) or T1+ARS (includes integrated bioinformatics module for automated secondary analysis with MGI’s workflows for WGS and WES).
How does the integrated DNB Make & Load module enhance workflow efficiency?
The T1+ features an integrated DNB M&L module enabling fully automated DNB preparation and dynamic lane loading within the machine. This eliminates the need for quantification and includes sample traceability. The reagent kit features pre-set design allowing one-click liquid addition without repeated manual handling. After extensive testing, the DNB M&L module achieves high stability (>95%) in making DNB, eliminating the need for DNB QC processes while retaining DNB for subsequent troubleshooting.
What are the flow cell specifications and performance capabilities?
The T1+ supports three flow cell types with varying throughput: FCS (500M effective reads, 2 lanes, 300G max output PE150), FCM (1000M effective reads, 2 lanes, 600G max output), and FCL (2000M effective reads, 4 lanes, 1200G max output). Run times vary by configuration: FCS PE150 takes 19H, FCM PE150 takes 19.5H, and FCL PE150 takes 24H. The system uses 500nm high-density chips with a pitch of 500nm for superior data output
How does T1+ handle third-party libraries and compatibility?
Currently, T1+ recommends using ssCirDNA or DNB inputs. For 5′-non-phosphorylated ILMN dsDNA libraries, manual conversion using MGIEasy Universal Library Conversion Kit (App-A) is required along with T1+App-D sequencing kit. The App-D kit contains Illumina TruSeq, Nextera, and MGI adapter primers supporting mixed detection. H2 2025 upgrades will add one-step OS reagent and T4 PNK enzyme for direct dsDNA input compatibility.
What are the data storage and analysis capabilities?
T1+RS version has 20TB hard disk space storing about 8 runs (PE150), or 12 runs with Q40 enabled. T1+ARS version has 40TB space. The system supports offline WFQ through UI interface and external WFQ capabilities. Automatic upload provides rsync upload and local mode through shared directories via samba and nfs methods. Default output is Q30 data, with Q40 available upon request through FSE activation.
What applications and sequencing strategies does T1+ support optimally?
T1+ is ideal for methylation sequencing, tumor sequencing, whole genome sequencing (WGS), whole exome sequencing (WES), and whole genome bisulfite sequencing (WGBS). It efficiently handles moderate sample sizes (10-50 tumor panel samples per run) with fast turnaround times. The system supports various read lengths: SE50, SE100, PE150, and PE300, with methylation throughput expected to reach 500M/lane with ECR1.0 upgrade in November.
How does T1+ manage reagent handling and maintenance requirements?
The system requires specific consumables including NaClO solution (Sigma 239305) for washing reagents, with Aladdin brand alternatives being added. The instrument needs 0.2mL 8-strip PCR tubes (AXYGEN PCR-0208-CP-C) for DNB reaction mixture, with 125 pieces included with instrument purchase. Four empty washing cartridges (PN: 940-002757-00) are provided for customer-prepared cleaning reagents.
What are the infrastructure and setup requirements for T1+?
T1+ requires a table supporting over 450kg weight (available as PN: 960-002351-00). The system currently does not support barcode-first reading (BBS) due to biochemical reagent limitations, but BBS functionality will be developed with future reagent upgrades. Different lanes on the same flow cell cannot pump different reagents – only different libraries or DNBs can be loaded per lane.
How does T1+ handle concurrent operations and workflow limitations?
T1+ARS cannot perform bioinformatics analysis while sequencing – once sequencing initiates on either side (A or B), bioinformatics analysis cannot start, and vice versa. For simultaneous analysis during sequencing, external bioinformatics tools like PFI should be considered. The system supports barcode balancing requirements and offers high process visualization with built-in instructional videos to eliminate operational concerns.
