T7+ - Frequently Asked Questions
What does “14T+ in 24h” mean?
The T7+ can generate over 14Tb of deliverable data within 24 hours, completing the full workflow from DNB preparation to cal file output within the same timeframe.
Do I need extra equipment for T7+?
No. T7+ features an integrated DNB M&L module that performs fully automated DNB preparation. Customers can also choose to prepare DNBs manually if preferred.
Can T7+ perform WFQ, and are there compatible platforms?
Yes. T7+ can run WFQ locally on its onboard server, which takes about 30 minutes per flow cell. Alternatively, remote WFQ can be executed using customer servers or Ztron Pro/Lite.
What sample types and sequencing strategies does T7+ support?
T7+ supports multiple sample types, including balanced libraries such as WGS and methylation libraries. Methylation libraries can be sequenced independently or mixed with balanced libraries, with the WGBS proportion ≤50%. Libraries from different applications can be pooled according to recommended ratios in the user manual. Sequencing modes include barcode-first, read1-read2-barcode, or custom strategies. Tests show no difference in quality between barcode-first and conventional sequencing, but barcode-first improves library splitting for short insert fragments.
How should reagents be stored and handled?
Sequencing reagent cartridges transported at ambient temperature can be moved directly to a 4°C refrigerator for sequencing preparation, while other reagents must be fully thawed before use. Some reagents, such as MDA enzyme, require frozen transport. Built-in containers are standard, but an external waste container can be connected if desired. Connecting an external pure water container is not necessary.
What is the workflow timing for DNB prep, cleaning, and WFQ?
Each flow cell has an independent DNB preparation module, and the process takes approximately 24 minutes per flow cell. Automatic post-run cleaning and reagent cartridge emptying take about 45 minutes. WFQ for a single flow cell takes around 30 minutes, and for four flow cells, about 120 minutes. WFQ can run in parallel with cleaning, DNB preparation, and DNB loading for up to three flow cells.
What happens if maintenance is neglected or liquid enters the needle wash port?
Neglecting maintenance washes may compromise sequencing quality, so regular cleaning is recommended. If liquid enters the YZ needle wash port of the sequencing reagent cartridge, drain all liquid, rinse repeatedly with purified water, and ensure the port is completely dry and free of particles.
How does T7+ manage data storage, compression, and failures?
T7+ has 62T of on-device storage. If running WFQ locally, it is recommended to store about two runs; for remote WFQ, about four runs can be stored. On-board data compression is supported, but decompression software must be installed separately. In case of Cal file upload failures or sequencing interruptions, manual backup may be required. Recovery functions work only if the server remains connected. The T7+ outputs double the data compared to the T7 despite having a similar flow cell size because reaction sites are more densely packed.
