The MGISP-100 Automated Sample Preparation System enables high-throughput sequencing library preparation by integrating all library preparation functions in a single platform. It supports automated operation of over 40 MGI and third-party library preparation kits.
With the MGISP-100, the MGIEasy Universal Library Conversion Kit (App-A) converts linear dsDNA libraries from third-party kits into single-stranded circular (ssCir) DNA libraries compatible with MGI’s sequencing platform using two steps: library adapter conversion and sequencing primer replacement. Combined with the automated Make DNB process, DNA nanoballs (DNBs) can be directly sequenced on the DNBSEQ™ platform.
The DNBSEQ™ platform’s unique Rolling Circle Amplification (RCA) technology avoids exponential amplification errors typical of PCR while reducing index hopping and amplification bias. Preparing the circular library involves enzyme cleavage, end repair, adapter ligation, product purification, PCR amplification, and circular DNA digestion. Libraries prepared on MGISP-100 shorten experimental time and improve stability compared with manual preparation.
Automation scripts for App-A+Make DNB on MGISP-100 have been extensively validated to ensure stability and repeatability and enable non-DNBSEQ™ libraries to be compatible with the DNBSEQ™ platform.
Automated library conversion includes four steps: PCR library preparation, automated pooling, circular DNA digestion, and Make DNB. It supports up to 16 samples at a time, taking 4.5–5 hours in total, with the operator performing only simple quality control for around 0.5 hours. Manual operation would require over 2 hours per sample.
Results show that MGISP-100 matches or exceeds manual preparation in AC-PCR product yield and library quantity. MGISP-100 with the APP series enables rapid, non-destructive conversion of linear libraries for DNBSEQ™ sequencing, enhancing platform compatibility, sequencing options, and system stability.
