(“MGI”), a company committed to building core tools and technologies that drive innovation in life sciences, today launched the DNBSEQ-G400RS FluoXpert in the APAC region and opened early access to the platform at Medlab Asia & Asia Health 2024. Integrating high-throughput sequencing with multiplex immunofluorescence (mIF) staining, the DNBSEQ-G400RS FluoXpert challenges conventions and pushes the boundaries of life science and technology as a multi-functional sequencer compatible with in vivo spatial protein detection.

Today, we are thrilled to introduce our gradable multiplex spatial phenotyping tool, designed to enable spatial biomarker discovery and revolutionise immunotherapeutic research.”
Supported by MGI’s DNBSEQ™ technology and a highly efficient mIF staining method, the DNBSEQ-G400RS can be upgraded via software to the DNBSEQ-G400RS FluoXpert, allowing users to obtain both molecular and cellular morphological data on the same platform. Users can seamlessly switch between sequencing and staining, achieving high efficiency. Its multi-task capability makes it suitable for applications including tumour classification, disease prognosis, tissue microenvironment analysis, and more.
Designed with systemisation and modularisation in mind, the DNBSEQ-G400RS FluoXpert integrates optimised hardware, advanced biochemistry, and innovative software to deliver an all-in-one multi-omics experience. The versatile benchtop sequencer features optimised optical, mechanical, fluidics, and electronic systems, enabling a faster and more streamlined sequencing process.
The platform also includes the user-friendly and highly flexible FluoXpert mIF reagent set. Compatible with frozen and FFPE tissue samples as well as off-the-shelf antibodies, it eliminates the need for complex conjugation. Available in standard and customised formats, the reagents incorporate efficient autofluorescence quenching and mild elution technology, ensuring minimal residual signal and optimal antigen preservation.
The FluoXpert programme supports fully automated fluorescence staining, high-resolution scanning, and imaging. With highly customisable protocols, it offers real-time image visualisation, antibody library management, and independent wash and elution. Images produced are high quality, with seamless stitching and 20x subcellular resolution (360 nm per pixel). Thanks to efficient fluid exchange and gentle elution, tissue integrity is maintained across multiple staining rounds, delivering robust reproducibility.
At 50 minutes per cycle, DNBSEQ-G400RS FluoXpert is three times faster than conventional manual staining and imaging, making it ideal for multiple staining cycles. Depending on flow cell choice, throughput ranges from 20 to 40 sections per week (based on an 85 schedule) with the standard 6-plex kit, reaching up to 140 sections per week (247) at maximum capacity. Results show no significant difference between mIF and the gold standard IHC, demonstrating excellent spatial protein identification performance and establishing the platform as a strong tool for proteomics research.

Leading with the question, “What If you could DIY your own innovative multi-omics tools?”, the initiative supports researchers globally in developing and implementing customised workflows for spatial transcriptomics, multiplex immunofluorescence (mIF), and other multi-omics analyses on DNBSEQ-G400RS. APAC researchers, particularly in Australia, Singapore, and Japan, can apply here before 30 August 2024.
